Study questions

  1. Compare cDNA sequencing and whole genome sequencing approaches: what’re their advantages and limitations? What information can a genome sequencing project provide as opposed to a cDNA sequencing project?
  2. Describe the chain termination procedure/Sanger method for sequencing.
  3. What’s the advantage of using modified Taq polymerase in DNA sequencing over the other polymerases employed in the old time?
  4. In the new sequencing methods, explain the principles and potential applications of the 454 pyrosequencing and Illumina’s sequencing by synthesis (SBS) method for genome sequencing. What’re their pros and cons?
  5. Describe the differences between hierarchical sequencing and shotgun sequencing. If you have a 700kb genome you would like to sequence, which of the two methods would you prefer to use and why do you prefer to use this method over the other?
