Study questions
- Compare cDNA sequencing and whole genome sequencing
approaches: what’re their advantages and limitations? What information
can a genome sequencing project provide as opposed to a cDNA sequencing
project?
- Describe the chain termination procedure/Sanger
method for sequencing.
- What’s the advantage of using modified
Taq polymerase in DNA sequencing over the other polymerases employed
in the old time?
- In the new sequencing methods, explain the principles
and potential applications of the 454 pyrosequencing and Illumina’s
sequencing by synthesis (SBS) method for genome sequencing. What’re
their pros and cons?
- Describe the differences between hierarchical
sequencing and shotgun sequencing. If you have a 700kb genome you would
like to sequence, which of the two methods would you prefer to use and
why do you prefer to use this method over the other?
